Callus culture for biomass production of milk thistle as a potential source of milk clotting peptidases Electron. J. Biotechnol.
Cimino,Cecilia; Vairo Cavalli,Sandra; Spina,Francisco; Natalucci,Claudia; Priolo,Nora.
The objective of this work was the optimization of the conditions of in vitro culture for callus production of Silybum marianum (L.) Gaertn. (Asteraceae). Sections of cotyledons, previously disinfected by washing successively with ethanol 70º, NaClO (10% w/v) and Tween 20 (0.05% v/v) and rinsing with sterile distilled water, were used as explants. For its initial culture, B5 medium supplemented with BA and 2,4-D solidified with phytagel was used, and a 63% survival was achieved. To obtain callus, two solid media were assayed (S1 and S2) using B5 medium supplemented with growth regulators (BA and 2,4-D or NAA and BA, respectively). The calli were grown at 25ºC during 45 days in darkness. Growth kinetics was studied using S1 medium...

Clonal propagation of the avocado: effects of the rooting step on graft union formation and development Ciencia e Investigación Agraria
Estay,Carmen; Cautín,Ricardo; Neaman,Alexander; Castro,Monica.
The etiolation and layering technique that is used on avocados has two critical steps, namely, the rooting of clonal rootstock and graft union formation between the commercial variety and clonal rootstock. This study analyzed the rooting step of the clonal rootstock regarding its influence on graft union formation and development. The optimum times for grafting the commercial variety onto clonal avocado rootstock were determined under two greenhouse conditions, heating and no heating. The results showed that under greenhouse conditions without heating, there was higher survival (%) in plants that were grafted 60 days after the rooting steps in the clonal scion, and, in the experiments in a heated greenhouse, the highest survival rate (%) was observed in...

Controlled Rhizogenesis and Mycorrhization of Hazelnut (Corylus avellana L.) Cuttings with Black Truffle (Tuber melanosporum Vitt.) Chilean J. Agric. Res.
Santelices,Rómulo; Palfner,Götz.
Hazelnut (Corylus avellana L.) is commonly used as mycorrhizal host for Tuber melanosporum Vitt. (black truffle). Black truffle orchards have been established in and outside its natural distribution area, currently also in Chile. Hazelnut can be propagated rapidly by cuttings, which, however, may not easily form mycorrhizal symbiosis under laboratory conditions. We successfully inoculated hazelnut cuttings with T. melanosporum and studied the effects of culture conditions, plant material and inoculum age on rhizogenesis and mycorrhization. Cuttings from sucker shoots showed the highest rooting rate (88.5%). Perlite and vermiculite as substrate increased root length (9.6 cm ). Treatment with auxin significantly increased root yield (up to 27 roots per...

It is possible to determine the optimum time for permanence of vegetative propagules (mini-cuttings) inside a greenhouse for rooting, and this value can be used to optimize the structure of the nursery. The aim of this study was to determine the dynamics of adventitious rooting in mini-cuttings of three clones of Eucalyptus benthamii x Eucalyptus dunnii. Sprouts of H12, H19 and H20 clones were collected from mini-stumps that were planted in gutters containing sand and grown in a semi-hydroponic system. The basal region of the mini-cuttings was immersed in 2,000 mg L-1 indole-3-butyric acid (IBA) solution for 10 seconds. The rooting percentage of the mini-cuttings, the total length of the root system and the rooting rate per mini-cutting were also evaluated...

Mini-incubators improve the adventitious rooting performance of Corymbia and Eucalyptus microcuttings according to the environment in which they are conditioned Anais da ABC (AABC)
BRONDANI,GILVANO E.; OLIVEIRA,LEANDRO S. DE; KONZEN,ENÉAS R.; SILVA,ANDRÉ L.L. DA; COSTA,JEFFERSON L..
ABSTRACT We addressed a major challenge in the in vitro clonal propagation of Corymbia citriodora, Eucalyptus urophylla and E. benthamii by using an ex vitro adventitious rooting strategy in a mini-incubator. Mini-incubators were placed in four environments for rooting. A shade house with no fogging system and a greenhouse with no ventilation but with a fogging environment had the best performance in terms of rooting, root growth and survival of microcuttings. Daily recording of the temperature within each mini-incubator in each environment allowed the verification of negative correlations between the maximum average temperature and the survival, adventitious rooting and root growth. The ideal maximum air temperature for the efficient production of clonal...

Rooting Platanus (Platanus acerifolia (Aiton) Willd.) cuttings in Marechal Cândido Rondon - PR, Brazil: Influence of lesions at cutting bases and depth of planting Acta Agron. (Palmira)
Rosa,Danimar Dalla; Tres,Gessika; Broti-Rissato,Bruna; Lorenzetti,Eloisa; Guimarães,Vandeir Francisco; Feiden,Armin.
Abstract Platanus, an arboreal and deciduous plant, is widely adapted and can be used for several purposes. Despite producing viable seeds, production of platanus seedlings usually occurs through vegetative propagation; cuttings are the best and most efficient source for obtaining seedlings. Although cuttings offer a practical and easy method to obtain seedlings in different vegetable species, they are influenced by several factors, both external and internal. The present study aimed to analyze the behavior of plantain cuttings planted in sand subjected to damage or no damage at the cuttings base, and planted at depths of 20 and 40 cm. Experiment was carried out at the experimental station of horticulture and protected cultivation of UNIOESTE-Brazil, in a...

Vegetative propagation of adult Ilex paraguariensis trees through epicormic shoots - doi: 10.4025/actasciagron.v35i1.15958 Agronomy
Wendling, Ivar; Empresa Brasileira de Pesquisa Agropecuária; Brondani, Gilvano Ebling; Universidade Federal de Mato Grosso; Biassio, Adriana de; Universidade Federal do Paraná; Dutra, Leonardo Ferreira; Empresa Brasileira de Pesquisa Agropecuária.
The difficulty and length of time required for seed germination of mate (Ilex paraguariensis), as well as the pressing need for clonal multiplication of improved genetic material, has resulted in several studies related to vegetative propagation in an effort to obtain rooted cuttings more quickly and with better genetic quality. Currently, the biggest challenge is propagating and rooting adult plants selected in the field without requiring clear cutting to generate conditions for the basal induction of juvenile sprouts. Therefore, the objective of this study was to develop a method to rescue adult mate plants through the generation of epicormic sprouts. To accomplish this, tree branches of mate that were at least 19 years of age were collected and packed...

Xyloglucans from Hymenaea courbaril var. stilbocarpa seeds affect Arabidopsis thaliana seedling growth by enhancing lateral root development Rev. Bras. Bot.
Salamoni,Adriana Tourinho; Sierakowski,Maria-Rita; El Jaziri,Mondher; Quoirin,Marguerite.
The effect of crude xyloglucan (XG) preparations from jatobá (Hymenaea courbaril var. stilbocarpa (Hayne) Y. T. Lee & Langenh.) seeds on Arabidopsis thaliana (L.) Heynh. root system development was investigated. The XG extracts exerted a dual effect on root system development by slowing down root growth and improving lateral root formation. These observed morphological changes were not due to oligosaccharides that could be generated following hydrolysis of the XG polymers, since XG hydrolysate induced a drastic inhibition of the overall growth process of the Arabidopsis thaliana seedlings. Histochemical test of GUS gene expression assay performed on seven and 14-days-old transgenic Arabidopsis thaliana plants carrying the CycB1;1-GUS fusion...
